Localized Topological Changes of the Plasma Membrane upon Exocytosis Visualized by Polarized Tirfm Jcb: Correction Jcb: Article

چکیده
In the discussion of the polar coordinate system leading to Eq. (1), the azimuthal angle f is ambiguously defined as " measured around the z axis. " A more conventional precise definition is " measured around the z axis starting from the x axis. " With this standard definition, the expression for S in Eq. (1) should contain the factor sin 2 f instead of cos 2 f. (The factor cos 2 f is correct only if f is understood to be measured from the y axis.) All of the geometrical models considered in the paper are azimuthally isotropic, so the starting axis for measuring f makes no difference whatsoever in the subsequent analysis. Also, in the equation for orientational distribution h(plane), shown before Eq. (5), the delta function factor d(u 2 p/2) is best written as d(cosu). This makes no difference for the particular case here, but a d-function distribution at any general polar angle u o must be written as d(cosu 2 cosu o).

Localized topological changes of the plasma membrane upon exocytosis visualized by polarized TIRFM
Total internal reflection fluorescence microscopy (TIRFM) images the plasma membrane-cytosol interface and has allowed insights into the behavior of individual secretory granules before and during exocytosis. Much less is known about the dynamics of the other partner in exocytosis, the plasma membrane. In this study, we report the implementation of a TIRFM-based polarization technique to detect...
متن کاملThe immunological synapse: a focal point for endocytosis and exocytosis
There are many different cells in the immune system. To mount an effective immune response, they need to communicate with each other. One way in which this is done is by the formation of immunological synapses between cells. Recent developments show that the immune synapse serves as a focal point for exocytosis and endocytosis, directed by centrosomal docking at the plasma membrane. In this res...
متن کاملA novel Netrin-1–sensitive mechanism promotes local SNARE-mediated exocytosis during axon branching
Developmental axon branching dramatically increases synaptic capacity and neuronal surface area. Netrin-1 promotes branching and synaptogenesis, but the mechanism by which Netrin-1 stimulates plasma membrane expansion is unknown. We demonstrate that SNARE-mediated exocytosis is a prerequisite for axon branching and identify the E3 ubiquitin ligase TRIM9 as a critical catalytic link between Netr...
متن کاملMembrane association and functional regulation of Sec3 by phospholipids and Cdc42
The exocyst is an octameric protein complex implicated in tethering post-Golgi secretory vesicles at the plasma membrane in preparation for fusion. However, it is not clear how the exocyst is targeted to and physically associates with specific domains of the plasma membrane and how its functions are regulated at those regions. We demonstrate that the N terminus of the exocyst component Sec3 dir...
متن کاملRho-GTPase–dependent filamentous actin dynamics coordinate vesicle targeting and exocytosis during tip growth
The dynamic activity of tip-localized filamentous actin (F-actin) in pollen tubes is controlled by counteracting RIC4 and RIC3 pathways downstream of the ROP1 guanosine triphosphatase promoting actin assembly and disassembly, respectively. We show here that ROP1 activation is required for both the polar accumulation and the exocytosis of vesicles at the plasma membrane apex. The apical accumula...
